This is one of the oldest "trick" questions commonly used in an Interview. It is a test to see how quick you are on your feet, how honest you are in your reply and how you handle a "difficult" question.
We all would like to change something about ourselves no doubt. I don't know many people that would not want to loose some weight or stop smoking etc.
However, you could really blow the interview if you said something like, I laugh a lot and I want to change that or I talk non-stop, or as some of these wonderful posters have said in jest I hope.
I not only have employed hundreds of people in my past, but I have interviewed thousands. I also have taught Interviewing Techniques and Resume Preparation courses.
I would tell them what I want to change most about myself is where I work. I would love to have a job where I was appreciated and rewarded for doing a great job, exceeding goals and for being reliable and responsible.

Aha!! "the" question...
the trick is to give them something positive about yourself, disguised as a negative.
I would say: " I can't think of anything really...maybe because I'm not afraid of hard work, I tend to take on a lot of work and end up doing too much."
or " maybe I tend to ask too many questions but it's because I really like to understand what is expected of me". |

I would say, and I have said, "let's cut the crap, I walk on water, so there's nothing for me to change, and if you didn't agree, I wouldn't be here talking to you right now." I got my fab job that I have now with that awesome attitude, I didn't even have to show any leg. That is smoove B. |

I used to answer: "oh, I don't stand up for myself sometimes" (totally bullshit).
BTW, I know every possible question an interviewer can ask. God knows how many job interviews I've done.
As for yourself, answer as follows:
"Well, you see, the thing I'd like to change....I'd like to see myself next year...with YOUR POSITION PAL!"

You want to be realistic, yet make sure it is something positive, and something you can work on, yet something that isn't required necessarily for the job at hand. Say they want someone that can 10-key. Then it isn't a good idea to suggest that wanting to learn 10-key is your biggest challenge.
If you are really crafty, you can make it sound like a strength, not a weakness.
Something like this: in my effort to get the job done right, I often find myself working extra hours. I would like to learn how to streamline my day better to get the job done right, yet provide time for additional tasks to be completed. I don't know...that's a weak example. But each interview will be unique, and each position or job skill being interviewed for may require its' own unique answer. |

Usually, in a work interview they ask what is your biggest defect.
I imagine it's almost the same question.
The answer could be: ''I am a perfectionist and this is what I don't like about myself.''
And u could add if u want: ''Everything I do has to be done till the end & in an impeccable way...'' |
