It'll probably take you about 3 days if you stick to the speed limit on the interstate hwys, which I strongly suggest you do because the hwy cops along there are absolute hawks with their hidden speed cameras so yeah prob about 2.5 - 3 days

The quickest way to Cairns from Melbourne is to go inland via Shepparton, West Wyalong, Dubbo, Walgett, Roma, Emerald, Charters Towers then onto Townsville and Cairns which is about 2950km or about 33 hours of driving. You could do this in 3 days
If you go from Melbourne to Cairns via Sydney and Brisbane, then it is about 3500km or 38 hours of driving. You could do this in 3 days but you'd be pushing it unless you are sharing the driving. It would take about 4 days to do this route if you are driving by yourself.
