From the Domestic Arrivals/Departures level of the airport, you'd want to catch the 424 to Airport Station. It's $2.50 initially.

Then, take the 98 B-Line Burrard Station which will take you to Downtown Vancouver directly.
Note: You'll have to pay an extra $1.75 to get to the Vancouver area.
Each ticket/proof of payment is required at all times when using public transit here and it's valid for an hour and thirty minutes!

Public transit is the cheapest, followed by the Airporter shuttle. If there are 2 or more people, a taxi is usually cheaper than the shuttle - the shuttle charges per person, whereas most taxis can carry 3 or 4 passengers and charge by the trip. If you are staying at a hotel in downtown, check to see if they have a complimentary shuttle service. Most of the Richmond and some of the South Vancouver hotels operate a free shuttle service.

Depends how many of you there are ... if you're in a small group (3-5 people) it'd be worth catching a cab.

OR go for the Airport Shuttle Bus, by memory I think it's about $20 one way. You can easily find the shuttle buses and the drivers and very friendly and knowledgable.

There is a new skytrain system under construction that will be completed next year, when it is it will not only be the cheapest but also the fastest as it goes above and below the traffic. Until then, check the busses.
