As long as your flight departs from US soil (Florida, New York or Puerto Rico, for instance) no. You just need a state-issued ID card or driver's license in order to board the plane (this also applies to the bi-weekly ferry from Fajardo, Puerto Rico).

If you are flying (or coming by ferry) from anywhere else (the British Virgin Islands, for instance), you must have a passport.

Are you an American citizen? Then, NO. The U.S. Virgin Islands are territories of the U.S. and you do not need a passport to travel to any state or territrory of the U.S. See site below and read sentence that starts with "NOTE" under the secton on "Land and Sea travel."

Whenever I travel, particularly flying I take a passport - aircraft can be diverted for many reasons so carrying your passport makes life a little easier. Most, if not all, airlines require some form of ID so once again its easier to take your passport.
