Hi Stephanie,
I'm just reading the other answers and see that nobody knows the Dominican Republic. OK ! to travel from Puerto Plata to Punta Cana by car, you will need about 6 1/2 hours if you are lucky in Santiago and Santo Domingo with the traffic jams.
The route is as fallowing :
Puerto Plata to Santiago ( 1hour)
Santiago (crossing the town) 1/2 hour
Santiago - Santo Domingo ( 2 hours)
Santo Domingo (crossing the town) 3/4 hour (if lucky)
Santo Domingo - La Romana ( 1 1/2 hour)
La Romana - Higuey ( 3/4 hour)
Higuey - Punta Cana ( 1 hour)

With public transportation (bus) it will take about all day! The reason is the timing of the buses. With Caribe tours, there is a bus to Santo Domingo every hour. Once arrived in the capital, you have to take one to Higuey. Arrived at Higuey, you need to take one to the village of Punta Cana and from there a taxi to the hotel/resort.
Here we are talking about 10 hours travel (minimum).
If you need to do this trip ask somebody with experience to do it with you. A taxi? way to expensive.
