The "best hotel" depends on what you want in a hotes. Do you want an all inclusive one or not? Other resorts cater to children while others do not allow children. We were there in July and our resort required age 14 and up. Some properties have great beaches while others have none. I suggest you decide what you want in a hotel and go from there. Then you can much more quickly narrow down which is the best for you.

The best hotel in Jamaica is in Negril.

RIU Tropical Bay in Negril. I definately think Negril is the most beautiful vacation spot in Jamaica. My wife and I went there on our HM and loved it. They were building a new RIU Negril as we were there. You may want to check it out.
