My suggestion, is to start here: http://cruisecritic.com/ports/index.cfm You can get some information on the destination before starting. This site is one of the best to look at as far as reviews and things for cruises.
Then, I would contact a travel agent to help you with the booking. Most of the time you will save money using a travel agent over an online discounter and you don't have as much risk. Travel agents often have lower rates than the cruise lines directly because they have group rates on some cruises. The best thing is to use one of the more well known agencies that specialize in cruises. I like using online travel agencies, but when I refer to an online travel agent, I do not mean Expedia, Orbitz, etc. I mean an actual travel agent.
As far as lines, I would recommend Royal Caribbean. I think the quality of the line is a little better than Carnival for just a little more, but in some cases it is a little less. Royal Caribbean also has great caribbean cruises.
