It depends on what you imply by "best way". The fastest is for sure to fly from Rome to Paris and then to La Rochelle. You could do the same for Bordeaux and Toulouse. By the way, Bordeaux is around 100 miles away from La Rochelle while Toulouse should be 300 miles from La Rochelle (not just nearby!). You could ride a train from Rome to Marseilles and then from Marseilles to la Rochelle with stops in Toulouse and Bordeaux (could take you one whole day). Or you could drive a car (2 days).

Because of the distance between the towns you will need a car but it is along drive from Rome. You can fly as the previous answer said

The train is your best bet on a budget. If you have more money, you can fly.
