Do you mean the Marais? Not that it makes much difference. Paris is very compact and everything is very easy to get to on the Metro.
If it was just one day I'd skip museums and see the five top "must see" tourist sights i.e The Eiffel Tower, the Arc de Triomphe, Notre Dame, Sacre Coeur/Montmartre and the Latin Quarter. I'd take a boat ride on the Seine, have a light lunch in a nice cafe and then a big classic French dinner and finish off the evening in a jazz club, drinking wine and listening to some tunes.
You could easily do all that in one day.
If there is just one thing then I'd say Notre Dame so that you could go inside and light a candle at the statue of the Blessed Virgin and say a little prayer that she would let you come back again when you had more time.
J'adore Paris
Definitely a visit to the Eiffel Tower.. It's an amazing structure. If you want to ascend it then get there VERY early to avoid the long queues. Otherwise Parc du Champs de Mars around it is a beautiful area anyway and so is the area aside the Seine. You will be able to take wonderful photo's.
Within a day I would recommend:
1. Eiffel Tower (early... open from 9am. Get there earlier!!)
2. Notre Dame
3. Sacre Coeur
4. La louvre (It takes a while to look at everything in there because it is so big!!!)
