Take the train. You can take an ICE (Inter City Express) in the direction of Basel, Switzerland that leaves every hour from the Frankfurt Airport. It takes just under an hour to Mannheim. It should be around 60 Euro roundtrip in 2nd class (this was my recent price to Karlsruhe which is just the next stop). You can purchase tickets in the Train Station in the basement of Terminal 1. They can also make seat reservations for a small extra fee, if the train is crowded or you want to make sure you get a seat.
