I know I have to go to the Vatican and Sistine Chapel and the Colosseum, but is there another 'can't miss' that's off the beaten path that not everyone knows about?...
Yes, you can. It takes 2 hr 45 min to 3 hr and you'll need to change trains in Florence. There are one to three departures from Rome per hour that make good connections. This includes some early departures and late returns that would give you a lot of time there. I often make day trips longer than this using the trains on the weekends when I'm not working here. You can check specific schedules and prices at the Italian train website (English version): http://www.trenitalia.com/en/index.html
