The best area in Tuscany to stay if you want to take trips to Venice or Rome is in Florence. Florence is a major hub for the trains and is on a more direct path to Venice/Rome than Pisa. Florence to Venice is about 3hrs and Florence to Rome is about 1.5hrs both by a Eurostar train.

The best option would be to rent an apartment in Florence. If you decide to stay in the Tuscan countryside or Siena, Lucca or Chianti then a car is necessary unless you are staying close to a train station - which most larger villages have one but some villas are not anywhere near a train station. By the way, if you stay in Florence, a car is of no use to you in Florence! The streets are narrow and it is difficult to find parking.

Rome would be a better daytrip than Venice since it is closer, but Venice is doable, it just would be a very long day since it is 3hrs one way to Venice. I recommend you see the Accademia and the Uffizi in Florence but you should pre-book tickets as the queues can be very long to get in - you can call 011 39 055 294 883 to make the reservations, they will give you a reservation number, just show it to the ticket box office at the respective museum.

Pisa airport has a railway line that runs to the centre of Florence. Both are in Tuscany. You can also catch a train from Florance to Rome

Florence is the major transportation hub for Tuscany, so if you rent a place that you to can get to Florence from, you should be set.

Florence is the most centered so the best for catching EuroStar(1/2 hour ride to both Venice and Rome) trains but the rent is insanely high. If price is not an issuse, than stay in Florence. If it is, than you could stay in Pisa, it has a train line but you would have to take a train to Florence first and then catch a decent train to Venice or Rome
