No they don't require you to have one but many offer them to you if you want to buy one. If you are planning to stay in alot of different hostels all over italy or europe , I would recommend you invest in getting one many times they give discounts.

I haven't stayed in a hostel for awhile and I have been living in Rome for the past 4 and a half years; yet, I do not remember if hostels require a special membership. It doesn't sound like you need to have one. Check out Hostel world, I went through them when I was traveling and I did not need a membership and they are a great service.
