first of all, greetings from Italy! my country, actually.
I see that evrybody suggest the most known towns, as Rome, Naples, Florence, Bologna, Venice, Milan, Turin. And that's correct, if you have no that much time, let's say only 10 days. If you have more time I would suggest to visit the surroundings of

Rome: Orvieto, Assisi, Gubbio and the so called Roman Castles
Naples: apart from Pompei and Capri (they are great) Salerno
Florence and Tuscany: San Gimignano, Siena, Volterra
Bologna: on the way to Milan I would stop at least in Modena, Parma & Fontanellato
Milan: Como and its lake, Vigevano with the square designed by Leonardo, Pavia and the Certosa. On the way to Venice I would stop at Bergamo, Garda's lake (example Sirmione), Verona (where Romeo and Juliet are from...) Padova
Turin: I would try a one day trip to Valle d'Aosta to have a look at Monte Bianco, monte Cervino and at some Castles over there
Venice: I would travel thru the Laguna to Chioggia
I suggest a handbook guide, of course.

It all depends on your interests. If you're in to history, I'd check out Rome and Pompeii first, they're very nice.
My personal favorite city on my trip to Italy was by far Florence, but Milan and Venice are also very nice.
I also HIGHLY recommend taking a trip out to the isle of Capri, it's really a sight to behold.
Also, I'd look into Assisi, it was by far the most pristine city I saw.
I never had the chance to go to Torino (Turin), but considering it was a housing place for the Olympics, I'm sure there's plenty to see there as well.

Depending of the time you have, you can go down this list up to the number of days you have (the number I put is what I think is the minimum to at least know the city in general, but I suggest, if you can, to stop even more if you like):

1) ROME: it's the capital city, the italian biggest one and probably the most full of history in the whole world. It needs no presentations - 4 days at least

2) VENICE: it must be visited for its peculiarity, there aren't in the world other cities built on the water. There are obviously a lot of places of interest in the city, but what really make the city worthy to be visited is the atmosphere. If you go there make a small trip to visit Murano and Burano islands, very peculiar too - 3 days

3) NAPLES: city worthy to be visited for his monuments, its castles and Vesuvio, but i'd go there expecially to eat the real pizza and to feel the tradition of the italian more traditional city - 2 days

4) FLORENCE: if you are particularly interested in arts and culture, you could stay there also a week, visiting Uffizi and a lot of other historical palaces, museums and church. If not it's a beautiful city to be visited, but not so big, so 2 days can be enough.

5) BOLOGNA: beautiful city with its two tower (they leans just like Pisa tower) and its porches (almost all the streets of ancient city have porches). It's not very big and if you are very hurry, one day could be enough to have a general idea of the city

6) MILAN: The most modern between italian biggest and most famous cities (it's in major part built in 19° century), so there are big palaces, skyscrapers, large streets and squares: it's the italian city that most seems like the other famous european cities, such as Paris, London, Vienna, but apart from the center, with the beautiful "duomo", Sforza castle and other, there is no much to see - 1-2 days

7) GENOA: It's undervalued from tourists, who usually go there only to see the acquarium (by the way, if you are interested in the undersea world, visit it, it's very exciting) and then go away. But Genoa has the biggest historical center between all european cities, even bigger than Rome one. Its house are very characteristic, very tall and close to each other. Spend one day there (two if you wanna see the acquarium) can be a good thing

8) PISA: Apart from the tower and the zone close to it, there is no much to see. You can stop there half a day while going to or coming from Florence

9) SIENA: Very beatiful city, quite small, but full of monuments and history. It's not far from Florence, so it could be easily reached sojourning in Florence

9)TURIN, PALERMO, VERONA, PADOVA, PARMA, LUCCA, PERUGIA are other cities you can visit if you stay really a lot in Italy

Apart from cities then there are a lot of beautiful regions, landscapes or small towns, like Valle d'Aosta (mountain region, with the Monte Bianco), Portofino and Cinque Terre (tipical little towns on the sea), Lake Maggiore, Lake Garda, Volterra (Tuscany town), Assisi, the "Reggia di Caserta" (an enormous palace like Versailles one), Capri (island near Naples), Pompei, Penisola Sorrentina, "Valle dei Templi",...

all Italian cities I've visited so far are great! Rome, Venice, Pisa, Florence, Milano, Verona, Sienna and many more.
You can go to Tuscany which is astonishing, rent a car and visit Orvietto, Sienna, Lucca, Florence, Montepulciano, Casole d'Elsa, Volterra, Pitigliano, Pisa, San Gimignano etc

Venice, Florence with a night in wine country near Siena for wine tasting and great food, Rome, and my favorite Capri with a visit to Pompeii and Positano. I wasnt really impressed with Milan and glad I was only there for 1 night, but I wish I made it to Lake Como while I was in Milan, it looks gorgeous. Verona is nice too.
