How can I get from Fiumicino Airport (Rome) to Vatican?

I'll be in Rome for 5 or 6 hours only and I'd like to get to see as much as possible.

You have a few options depending on what your actual time in Rome is and your budget. One option is to hire a car with a driver to take you on a tour of Rome including stopping at the Vatican. All the companies offer this, but it's expensive. If you mean you only have 5 or 6 hours from arrival to the next flight, this has the advantage of being most time efficient.

You can also take the Malpensa Express train from the airport into Rome. It arrives at Roma Termini train station - it's 11 euro each direction and takes about a half hour each direction. When you get to Roma Termini, you can get a cab to the Vatican if that's what you're specifically interested in. Another option is to take the hop-on, hop-off bus which takes a loop around the major tourist area - you could at least see some of Rome while you're there and decide where you want to get off and explore more. when you arrive in Fiumicino Airport you have to take the train "Leonardo Express" who will comes and goes from/to Stazione Termini (main train station in Rome).
Then you have to take subway Linea A and exit in Ottaviano/S. Pietro.
Then you have to walk for 10 minutes and you will be in Piazza San Pietro.

The Leonardo Train it's every 30 minutes (I ain't sure about it but almost) from 05.52 to 22.52 from Stazione Termini to Fiumicino airport and from 06.37 to 23.37 from Airport to Stazione Termini.
The Leonardo Express is a direct train, no stops in the middle.

if you have only 5 or 6 hours take a cap, no way you're going to make it in time by public transport! Try to see Saint Peter as well as the Angel's bridge, then ask the cab driver to drive you back passing by Piazza venezia and the Colosseum!
