Get a Euro rail pass for Italy before you leave the country.
The train station in Florence isn't far from the Duomo,
and the first class cars are very nice. We travelled by
train, with the Euro rail pass all you have to do is get to
the station and find the next train to Rome, for example,
and find a car with empty seats (tho that can be tricky
in the summer-we went in May and the cars were
pretty empty) In Rome you change to a train for Naples,
then to the train for Pompeii. Just be careful to get off
at the Old Pompeii station, we made the mistake of
getting off at the first station marked Pompeii and ended
up in the center of the modern town, so stay on until the
Old Pompeii station, the ruins are just across the street.
Make sure that you get there early as the ruins are quite large
and there are lots to see and you can run out of time if you
come in the afternoon. Pompeii is incredible, one of the
places in the world you really need to see.

If you like to fly, then the closest airport to Pompei is Naples Capodichino (NAP). However, there's no direct flight from Florence (FLR) to NAP. You have to connect through Rome using Alitalia flights. I made a random search and found the lowest rate to be around US$294 for a return flight. The rate might differ for the days you want.

By train, you can travel from Florence to Pompei by connecting in Naples. Again, I entered random dates and got the lowest fare at US$79 for a round trip.

I could not find a specific site for bus services, but I'm sure there's something cheap that can take you on this trip.

By car, the trup takes about 4 hours and 15 minutes. You have to take Route A1 heading south. Shortly before reaching Naples, you should turn on Route A30 to get to Pompei.

I agree with the first persons answer regarding the train from Florence. I'd also definitely suggest taking the first class rail to Naples. You will enjoy this. Also, once you arrive in Pompeii,
make sure you pay a private guide to escort your through the ruins. Just make sure he is a real guide. Having a guide is very worth it and will enhance your experience immensely.
I'd say that seeing the ruins without a guide is not worth your time. I paided about 50 Euro for a private guide when
I was there. Also, if you don't mind paying more money, I had a driver pick me up from the Naples train station and
tour me around the Amalfi Coast and take us to Pompeii, where I picked up another private guide to take us
Into the ruins. You will get to see so much with a guide/driver for a day. This way your not traveling the whole day via Train. You will be in a car where you can stop whenever you feel like it. A travel agent or a concierge service can
arrange all of this ahead of time.

I was based in Rome and did a day trip to Pompeii. This included bus trip, tour guide, lunch and entry into pompeii. There was option to see capri too. I was travelling by myself so this was a good option
