I spent 23 hours there in 2003, it was enough to whet my appetite, but I'm dying to go back for at least a week. If you want to concentrate on one area, a foot tour might be best, but I used the subway and got glimpses of as many different places as possible, the colosseum, the vatican, circus maximus, spanish steps, and a few others. You can buy a day pass on their system, I think it was about 8 euros.

Impossible task -- use local Metro bus; hop on and off. Don't get stuck on a tour bus. Make a list of must sees, such as coliseum, pantheon, etc. Try to get to those, but stop for a while and enjoy espresso or capuccino while watching the "people parade" -- that's the essence of Italia!

foot and taxi, Rome is full of so many great things to see. try to make a list of what you really don't want to miss. you can walk from block to block and see roman ruins. it's an incredible place to visit.

That is an impossible task. The closes you can come to achieving this would be to utilize the subway as much as possible. The best map I have found in Rome is at the information booth at the main train station, and the book DK Eyewitness Travel Guides are the best to help you figure out what you want to see in that short time; they are worth it. If you have the book Lets Go Europe, you have wasted your money. Never use tour guides there, you can find the information needed on the Internet to what you are looking at, and most of the time, they are just translating what the pamphlets are telling you and I have heard many say things that are out right incorrect. Emersion in Italian is a computer program that can teach you the needed language to get around quickly and efficiently.

Other info that may help you achieve your goal is that if you here someone say hello to you in your language, just remember, they are not going to speak to you unless they are attempting to sell you something that you most likely don’t want. There are individuals that are dressed like roman soldiers around the city as part of the attraction…ignoring them is the best policy. The reason I went into detail is because if you have just one day, you do not want to be delayed by tourist traps. If you have any other questions use the CCW, I lived in Italy for about two years and found myself in Rome a lot.

Spend more than a day.

I was in Rome for three days and saw everything I wanted to see in two days. Here's how:

On the first day, I bought an all-day Metro ticket and went from station to station looking at all the nearby sites. The Vatican, Colosseum, Trevi Fountain, etc. all the big touristy sites can be seen this way for less than 10 Euros this way.

On the second day, I bought a ticket for one of the hop-on/hop-off double decker bus tours. This is a little more pricey, but it'll get you around the most populated parts of the city the quickest. Buses are always going. As long as it's on a weekday, you won't spend more than five minutes waiting for the next one. If you really are going to only spend one day in Rome, this would be your best bet. You can get on and off at any stop and ride all day. Something like this:

On the third day, I had already seen everything, so I revisited a lot of the sites and did some last minute shopping.

I went there last winter and spent two days. It would take a long time to see everything. We went to the main attraction (colloseum, forum, Piazza Navona, etc.) We did most of it on foot, and believe me it was a long day. I hike quite a bit (did an 80 miler this summer) and I was still a little sore, but it was well worth it. Take LOTS of pictures.

P.S. don't rely on the bus system, very unreliable

ASRoma
You would have to go by tour for any attractions. Pick the most important to you for you will miss many! Rome wasn't built in a day and to see it in a day would be impossible! Due to traffic, half the day would be spent watching Vespa's fly by you at the traffic light's. Good Luck!
