Definately the Uffizi - it's wonderful, but make sure to take your time. Also, look into the Bargello (I think that's how it's spelled). Look at the Duomo and the Baptistry, as well as the many "squares" with art works displayed. Find an open air market and buy some fresh produce, bread and cheese and take a pic nic by the river arno!

Places not to miss:

The Bargello - it's often overlooked and not too crowded, but it's packed with beautiful sculpture by Donatello, Michelangelo, and others.

Santa Croce - beautiful church that's also not too crowded. The piazza outside is often home to fairs, and in summer you can sometimes watch a pick-up game of soccer there as well.

San Marco - this is a monastery which is decorated with frescoes by Fra Angelico - very peaceful and serene. Absolutely beautiful.

La Specola - a medical museum packed with wax anatomical models. Think Philly's Mutter Museum but even crazier.

Also, try stopping by the Central Market (Mercato Centrale), near San Lorenzo market - there are a few good places to eat inside on the first floor. The market is something not to miss as well. The Medici Chapel is reached by going to the back of the market and entering an easy-to-miss doorway on the back of the church.

For more tourist-packed areas, there's always the Baptistry and Duomo - remember that many of the sculptures you see outside are replicas. Go to the nearby Opera del Duomo museum (I think it's on Via Proconsolo across from the cathedral's apse) to see the originals, as well as some pretty impressive replicas of the machines used to build the dome. Also, check out the Ponte Vecchio - go down to the next bridge for a good photo op. Also, early morning trips to the Arno will sometimes give you a good glimpse of the so-called Arno river rats swimming in the current.

Oh, while you're at the Uffizi - stop for a hot chocolate. I forget the name of the resturant, but it's on the piazza across from the Palazzo Vecchio - the best in Florence (and probably the most expensive, but still worth it).

If you have time, take a bus up to Fiesole for a gorgeous sunset view of the city. Buy a ticket at any of the tobacco shops and make sure to validate it once you get on the bus - the fine is 50 Euros if you don't! There's some good places to eat here as well.

And one more tip: if you get attacked by mosquitos, stop in any local pharmacy (look for the neon cross) and get a tube of Fargan. I swore by the stuff when I lived there!

Hey Florence has alot to offer, depends what you are into. But you definetely have to see Ponte Vecchio, the Duomo (visit JJ Cathedral the bar) and Fiesole (u can take a bus there, 20 min).

Anyways, if you are looking for good bars then go to JJ Cathedral next to the duomo (lorenzo is really nice) or Shot Cafe if you want more of an italian atmosphere.

Good Discos are YAB, Maracana, or even Space Electronic. Also free bar/club which is nice is Twice.

Anyways, their is alot to see in florence but u are able to see it in one day if u invest the whole day. Go to all the Piazzas and take lots of pictures. Also MUST GO TO MICHELANGELO..you get the whole view of firenze, its walking distance from ponte vecchio-like 15min. Or u can take a bus or taxi there, just say MICHELANGELO...its beautiful!!!

Orsanmichele (impressive gothic church)
SS.Apostoli (medieval church)
Santa Felicita (wonderful frescoes by Pontormo)

All the above free, centrally located, and not so touristy :)

and Palazzo Medici Ricciardi chapel (a jewel with frescoed by Benozzo Gozzoli)

You have some really good answers here . I would just like to add ; as suggested a trip to Fiesole is a lovely fresh air change from the city ; it is only 7km away , about 1000ft high with spectacular views .
From Piazza San Marco , take a No . 7 bus ; you must buy the tickets beforehand at any Tobacconist . A 4 journey ticket is quite handy . Put the end of your ticket in the franching machine on the bus .
You can book a time for the Accademia , tel. 055-2388609 . Palazzo Vecchio and Palazzo Pitti usually no waiting , both interesting , Palazzo Pitti has the Galleria Palatina and Modern Art , the latter open in the morning only . Outside are the Boboli Gardens .

Watch the sunset from the Piazza Michelangelo. Some of my favorite pictures are from here. Florence is a lovely city to walk around. Lots of stores aroune Santa Croce church. There are some interesting people buried in the church, see below. Buon Viaggio!

Florence is wonderful - I'm surprised no one has mentioned a visit to the Accademia where the statue of David is housed (he is enormous!). You can also walk across the Ponte Vechio and keep walking until you reach the Pallazzo Pitti - home of the Medici family.
Santa Croce is worth a visit, Machiavelli, Gallileo and Michelangelo are entombed there.
Have a coffee in Piazza della Signoria and take in the magnificant history of the place!

Oh...btw, take a walk around San Lorenzo and check out all the leather stores...remember to smile a lot and do a bit of haggling!

The dome called Santa Maria Novella, the pitti palace the uffizi museum (even if you will always find a long queue) signoria's square, the vecchio ("old") bridge
Have a nice holiday..I as italian and from florence say that it's a beautiful and unique city because in the word you will never find a place so rich of story and culture
