If you can walk a few miles each day, I recommend the following

Galleries:
Uffizi (make reservations or WAIT)
Accademia

Churches:
Duomo

Points of interest:
The Ponte Vecchio
Fountain of Neptune
Boboli Gardens

Shopping
San Lorenzo Leather Market

Avoid the big fancy restaurants and stick with the little places down side streets. The food is great everywhere and the ice cream is beyond belief but fattening! There is a pizza shop near the leather market that must have 100 types of pizza. Ever see octopus or eight cheese pizza? Sport bars can get you a coffee fix to pick you up.

The hotels with a river view are more expensive than those further out, and are not worth it. The Arno is like the Hudson River in NYC.

The other answers are quite good. I'd only add that if you like gelato then you MUST go to Vivoli's. I've been to Florence a good half dozen times and after my first few visits my friend (who lived there for a year) kept after me... "Why didn't you go to Vivoli's??" When I finally went I regretted not going sooner. The gelato is to die for! Far and away the best I've ever had - and I used to summer in Italy and visited a fair number of cities.

In addition to what people have already said, I would say visit Santa Croce. You will not have to wait in line to go in and it is absolutely beautiful inside! There you will see the tombs of Michelangelo, Galileo, Dante, and many more... Make a reservation for Accad. and Uffizi because the wait can be rediculous. When i went a few weeks ago the line to see David was 3 to 4 hours long in 100 (F) degree weather. Be sure to walk across the Ponte Vecchio one night, when you do I suggest going to The Carraia gelateria which is two bridges down from the Ponte Vecchio on the south side of the Arno. If you are brave you can go sit on the bridge over the ledge. There are small triangular areas that people like to sit on right over the Arno!

go see David!!! he's huge lol!!! you can also just walk around and see all of the really cool scenery. When i went me and my family went to go see a famous Italian writer's house and it was SO cool and old!!! you'll have so much fun there:)

You can go to the Uffizi Museum, the Galleria Academia, you can explore the shops which have really unique things, you can go visit the cathedreals. Check www.frommers.com has list of every attraction with a brief description.
