Tuscany/ Umbria which towns to visit?

Planning a trip to Italy for next September. We want to visit Tuscany & Umbria, but only have 9 days to do so. We have read about Perugia, Assisi, Gubbio & Spello in Umbria and Siena, San Gimignano, Montepulciano, Lucca/ Barga & Cortona in Tuscany.
We plan on renting a car to have an easier time getting around. What I need to know is in 9 days which of these I could miss, if any and what route/ order would be the best to take to travel to these places?
Lastly considering we will probably be moving around a lot does it make sense for us to base ourselves somewhere where we can pre-book acommodation or to travel around and try to find somewhere on a whim? I figure if the worst happens we can always sleep in the car.
Additional Details
Flying into Venice and having 2 day there, then taking the train to Florence for a day or 2, then driving around Tuscany & Umbria then train to Rome for 2 days before flying out of Rome. Now you know where I'm going into/ out of is there any more you can tell me?

Any or all of those would be great places to go. Rather than trying to hit 9 cities in 9 days without a definite place to stay, I would have a base in Perugia to see the Umbrian towns - you can actually see Assisi from the upper city in Perugia. When you've seen the four towns there, move on to Siena and spend a day from there in San Gimignano and another in Montepulciano. You can stop in Cortona on the way to Siena. Depending on where you flying in and out, you could do the trip in reverse.

Personally, I like Lucca and Barga a lot, but you just don't have time to really do everything on your list in 9 days ... and you're totally missing Florence.

In my opinion you have to stay in Tuscany, its really beautiful region. Florence the city of Tuscany is well known for it's beauty.

From my point of view nothing could be missed but trying to do everything may be a little hard.
We had a week with my family in Tuscany and chose to stay in a villa close to Montecatini Terme, halfway between Florence and Lucca.
This gave us the chance to take the best from this trip even if we'll certain come back to visit Assisi and Gubbio as well.
Well, I suggest just to plan in advance and not getting there just trying to see all...

September is a beautiful time to visit Tuscany and Umbria: it is not too hot and busy, like in full summer. Renting a car is also a good choice. Since you are driving there, you should take into account that there is a 4 lane road going from Siena in the heart of Tuscany to Perugia and Assisi in the heart of Umbria. My recommendation is to choose a place between these two towns and make one or two day tours from there. This area happens to be one of the most beautiful in Tuscany, going from Siena, the Chianti area through the Crete Senesi, Montepulciano and Valdorcia, Valdichiana to Lake Trasimeno, Perugia and Assisi.
The A1 motorway and trains going from Florence to Rome also pass through this area: this makes it very easy to make even one day tours to both cities.
I would definitely visit Perugia and Assisi in Umbria, Cortona and Montepulciano, the Valdorcia, Siena and of course, Florence and Rome. I find San Gimignano too busy and the neighboring Volterra much more fascinating and true.
