I have been there all hours of the day but I also always go around late September early October, so it may be off season.
I also found this post that may help you, check out the link below. |

Hello,
If you don't want to do a guided tour, then get there after 12 or 12:30. The line dies down in the afternoon so going after lunch will enable you to skip the long lines you find in the morning. The line in the morning can be as long as two hours, with people having to go through security and then waiting in line for tickets.

My recommendation is get there at 6:30-7. You still might have to wait a while to get in, but not too long. Its worth it to get there early. And also the museum isn't as crowded if you get in right away which is nice. |

You will have to que regardless of what time you get there. No one can predict how many tourists will go. However bare in mind all the tour groups go in the morning, so perhaps going just as the doors open (about 8.30am) is better as they can all flock in.
Even if you do a guided tour you will have to wait in the line. I live and work in Rome for a tourist company and we sell these trips and our guests have to wait in line. Our trip gets them there at about 7.30 so they are waiting before the doors open. |

Having done the Vatican Museums recently, I would advise considering, taking a guided tour. Museum tickets 18 Euros, the tour including the tickets was 50 Euros. The regular line was 800 meters long (at 9:30). With the tour we spent maybe 15 minutes in line. Instead of spending half a day in line, we paid the extra 32 euros and go an extra half day of sightseeing in Rome.
If you think that the 32 euros is too steep, look at the numbers.
Take the cost of your stay in Rome, divide it by the number of days you have to actually see Rome (don't include arriving and departing days). That gives how much it is costing you per day to be in Rome. Do you want to spend a half a day waiting in line at that price, or would you rather be doing something else. |
