The trip could be rather shorter than previous answer's 3 and half hours. If you check on the below link with time table you can see that there are chances to make the whole trip in about 2 hours and half.

Take the train that leaves straight from Fiumicino Airport railway station to Roma Termini (half an hour trip). Here you can take any Eurostar train (reservation of seat is mandatory; you need to login to the site and then you'll be allowed to buy tickets and make reservations on line by using the same website).With Eurostar it will take further 2 hours from Rome to Florence.
That's the best way to travel in Italy (faster and cheaper).

There's a train that runs between Fiumicino and Termini station in Rome. It takes about 30 minutes. From Termini, you then transfer to the train going to Florence, which takes about 3 hours.

There is a train in the airport that will take you directly to the Termini. From there you can take a Eurostar train from Rome to Florence in about an hour. We were there in March and it was $66 for both of us.

I'd rent a car to be on the save side - their train schedules are worthless (own experiences over years).
