Take the 110 Trambus at the Termini. It is 13 Euro and you can get on and off all day. If you get on at 8:30 and ride it all day you should be able to see just about all the important things. Vatican City, Coloseum, Campo di Fiore, Pantheon, Spanish Steps, among other things.

Vatican and Coliseum are must sees but I would suggest signing up for a tour. You get to go past the extremely long lines which is a huge time saver, plus you learn so much more with a tour guide. You can join a tour right in front of the Coliseum so you don't have to be there at a specific time. Have a great time. It's a great city.
