I'll suggest to do an Open Bus Tour. In a day you'll see the main beauties of Rome, in a great hurry, but is all you can do.

yr visit to rome wil b incomplete if u missed any of it.the most u can do is to organise it intelligently.
reach vatican before 9 am and pay 35-45 euros for guided tour to visit the museum and the sistine chapel.the guided tour will save u 2 hours waiting time in the que.if u are ready to wait however,u neednt take it.the beauty of the sistine chapel and the museum are worth 2 hrs que.
you will be out by 1.30 appx. carry sandwiches or any fast food so that no time is lost in eating.now head straight towards the st.peters.que again but a small one.
visit the cupola after climbing 300 stairs to see the whole of rome from the top(worth the climb) or leave it if u wish and merely visit the cathedral/tombs of the popes.IT IS EXTREMELY GRAND AND BEAUTIFUL.
next come out of the basilica and hop on the open tour which will take through the whole city and even show u colloseum,palatine hill,tiber,ruins etc and drop u on metro station
i have just yesterday returned from rome and did exactly as i have written here.

I really only had one day in Rome, and this is what I did:

-the Vatican (get there early. Early. Leave when it's still dark outside, if you can. Everyone goes to the Vatican, so there can be huge lines). The Vatican museum has plenty of stuff to see, plus you get into the Sistine Chapel and St. Peters. Wave to the guards if you see them. They waved back to us.
-the Colosseum & the Forum, which are pretty much across the street from each other
-the Pantheon
-the Spanish Steps

I did them in that order, in one day. Take the bus as you need to. It's not always reliable, but it's cheap and goes where you need to. Invest in a map. Be prepared to walk. If you can, see if you can buy tickets for the Vatican museum and the Colosseum ahead of time. If you're going there in the summer, remember sunscreen and water. Be assertive when crossing streets - no one stops for you unless you're already in the middle of the road.

I would skip the museums, since you have only one day.
St Peter's Basilica,the Vatican, is worth the visit and like someone suggested, go right to the top and it's a great view! That's so true. Don't miss the Sistine chapel there, it's spectacular. It is a painting of the ceiling by Michelangelo.
The Trevi fountain is lovely too and a must see. The Colosseum and the Forum are must-sees too. Take it all in, in a relaxed way so you will not tire out. Because you have only one day, there is a great temptation to see as many things as you can. But I would say also that you want to enjoy every detail to remember for years to come. If you rush through, you may not get that full effect of enjoyment.

With only a day in Rome, I'd head for Vatican City. There's a ton to see, and you don't have to go far to see it. St. Peters, The Vatican Museum, etc...

If you only have one day then you should take that day and do as many little things as possible. When I mean little things I mean that you should see things that will not take all day. St. Peters doesn't take long to go through and I suggest paying the extra 10 or so dollars and taking the tour up to the very top of the cathedral. Not too many people take the trip but it only takes about 1/2 an hour to an hour and you get an awesome panoramic of Rome. I would suggest that you skip the museums because they are really packed and you do not want to spend most of you only day in Rome in a long line. You should stop by the colleseum and check out the forum as well and do not miss the Trevi fountain. Other than the colleseum, none of these should have lines especially not Trevi fountain and the forum which are pretty cool places to see and sort of sit back and experience. I hope you have an easy time deciding because it is hard to do I stayed 3 weeks in Rome and still didn't see everything. Good luck

I would definitely go to the Colosseum. It is right on top of the Roman Forum. Also, Colosseum tickets include admission to Palatine Hill, which is right beside the Forum.

Then I would head to St. Peter's Square. I would skip the museums; they will eat up your whole day. The Spanish Steps/Fountain of Trevi/Pantheon are reasonably close to each other, so you could see all of them, too. You might look into one of those bus tours to try to see as much as possible. You can see quite a lot if you are willing to move quickly.
