You can catch the grey bus to any of those locations from San Jose.

Please understand that the Bus Terminal is located in San Jose’s Coca Cola district, aka Zona Roja, or the Red Light District. Zona Roja is one of worst parts of San Jose for pick-pocketing and petty theft. Obviously, tourists and travelers are targets. Keep an eye on your bags and belongings at all times, and keep your passport and important stuff close to you. Don’t let anybody watch or handle your stuff for you.

In response to your second question; Tortuguero, Costa Rica is accessible only by boat or small plane.

Even though I live in Costa Rica and own a car, my preferred transportation to either coast is the wonderful Costa Rica bus system where approximately $7 will get you a ride on a full-sized modern bus with reclineable padded seats.

There are two bus+boat options for getting to Tortuguero...

Route A: 8 Hours (3 hours by bus + 5 hours by boat):
Starting in San José at the Caribbean Terminal (Terminal Caribe) you take a 3 hour bus ride to Limón. At the Puerto Limón terminal you will switch to a local bus headed to Moín at a fare of approximately $0.40 per person. At Puerto Moín you will take a boat for a $35 per person 5 hour, 50 mile ride up the Caribbean canals and rivers that lead to Tortuguero.

Route B: 4 Hours (3 hours by bus + 1 hour by boat):
Starting in San José at the Caribbean Terminal (Terminal Caribe) you take the bus through Guapiles to the Cariari Bus Terminal. At the Cariari terminal you will switch to a local bus going to La Pavona via a very bumpy gravel road that winds through banana plantations and other various types of farms or "fincas". At La Pavona you will need to take a water taxi for an approximate 1 hour boat ride through small canals at an approximate cost of $5.00.

I suggest taking one route to Tortuguero and the other route back to San José being as each is quite unique in what you will experience.

Be sure to check the source links below for more Tortuguero and Costa Rica bus information.
