The best way actually is to try to get some info from truckers that actually make the run from the US to Panama and beyond. There should be a website where you can find some info about it, or maybe they have a union where you can ask questions about the trip and the requisites you will need in order to properly cross your car through the several borders you have to cross.

As for safety, yes you do have to be very careful as you will be going through some trouble spots. Besides car theft is very common in Central America (excluding Panama). Try to make all your longest stretches during the day, and also try to cross the borders during the day in order to assure that there will be customs and immigrations officers. Some borders do close for business at night.

Call US Customs to check what do you need in order to take your car all the way to Panama. They can provide some help there. Get worldwide hospital insurance in case you need medical attention during your journey, and carry enough cash to get around, and keep the rest in your bank account. Get a list of ATM's around the world where you can get cash, or use credit cards that can get replaced anywhere in the world, but don't flash them around.

Buy some country guides in order to get a sense of what's to avoid and what you shouldn't miss. They provide great info about safe lodging and travel times & tips.
