Well, the first important city is Guatemala City (the capital), it's a city with all you may need.
The second city in importance is Quetzaltenango (also called Xelajú in indigenous language). It's a city that mixes the modern aspects with a beautiful historical center. This city is located 195 kms from Guatemala City.
But... we can't forget Antigua Guatemala, the colonial capital. This city is historic patrimony of the humanity. A quiet city where the time seemed to stop. It's only 30 minutes away from Guatemala City.

Guatemala City: the biggest city in Central America. The perfect mix between modern and colonial infrastructure.
Quetzaltenango: more than 1 million inhabitants. Located west to Guatemala city.

Though neither are technically cities as we know them I would say:
Antigua : former capitol...lots of history and culture.
Chichicastenango : has the largest market in the county on Thursday and Sunday(the whole town becomes a market) contributes a lot to the countries economy.
