There is the possibility that your passport will be checked MANY times while you are traveling to Mexico (I'm going to assume from the United States). Not all of these ALWAYS happen, but these are various points at which I have been asked for my passport on my way from the US to Mexico:
1. When checking bags at the airline counter in the airport. If you are printing out your tickets, oftentimes the attendant will ask to see your passport.
2. When going through security. You will need to show your boarding pass and an ID -- usually I already have my passport out and so use that again.
3. If TSA questions you about your belongings, you may be asked to show your passport again.
4. I have been asked (sporadically) to show my passport on an early leg of a multiple-leg flight even when that flight does not yet take me outside the country.
5. Before you board the flight that WILL take you into Mexico, Continental flight personnel ALWAYS check passports.
6. When you complete the paperwork for your tourist visa, you will need your passport to complete that and Mexican Customs and Immigration will most likely check the information on the form against your passport.
7. When entering Mexico, if your baggage is screened and checked, they may once again ask to see your passport.
It is wise to leave a photocopy of your US passport in a secure location before you travel. It is also recommended that you have a photocopy of your passport (or at least have copied all relevant information and numbers) with you when you travel and keep that separate from your actual passport.

Usually they check your passport two times when coming by plane from a foreign country to Mexico. The 1st time your passport will be checked on the departing airport and the 2nd time the Mexicans check your passport when leaving the transfer area.

You will go thru customs and immigration at the airport , and at that time they will stamp your passport. They also will give you your 6 month visa card - do not lose - you will need it when leaving Mexico and its a problem if you dont have it.
If flying into Mexico City - the old terminal is chaos in the customs line. It circles in and around the luggage carosal and you have no idea where the end of the line is :) Ah , the joy's of international travel.
