All routes on the south island are a pleasure to drive! The scenery is stunning and constantly changing.
The quickest route is definitely via Lake Tekapo - takes about 6 hours.
But if you have time (days) to kill I'd definitely take the long scenic route over Arthurs Pass and down the west coast past Franz Josef Glacier and Fox Glacier. On to Haast and then on to Queenstown via Wanaka. Then if you're heading back to Christchurch afterwards you can go back via Lake Tekapo for a shorter drive.

I'd also recommend driving yourself - there's not a lot of traffic and it gives you the freedom to stop off and explore all those interesting spots you'd normally shoot past. Car hire can be pretty cheap in NZ and you could always ask about "relocations" which are free (apart from the petrol).

From Chch go south on the main road (state highway 1) then turn inland at Geraldine (highway 79) through Fairlie (onto highway 8, the Lindis Pass) and through Twizel, then turn of it when you get to Cromwell. Go through Cromwell and follow the signs to Queenstown
