The easiest and most cost-effective (though not the fastest) to travel amongst major cities in Spain is the railway. You can take the bus too, but I'd suggest the rail. www.renfe.es
If you're in a hurry, and are willing to fork over some cash, Ryan Air is a great way to travel too. They're really cheap and reliable.. The only downside to Ryan Air is that they don't fly in and out of major airports like Madrid Barajas, and the main airport in Barcelona ... they fly to outlying areas and smaller airports ... it's like Europe's version of Southwest Airlines
