I think the cheapest and fastest way is by train, you can go by AVE ( Spanish faster train, like the French TGV ) from Sevilla or Cordoba to Madrid, and then a train to Salamanca.
Other options are by bus, but this is slower and not so comfortable, or, if you can drive, hiring a car, but may be more expensive.
Any case, you have to visit Salamanca.
Is one of the most beautiful towns of Spain, their ancient streets are full of art... and if you go between end-september and june you will find a lot of nightlife, amazing! Because Salamanca has one of the most important Universitys of Europe. |

I'm doing that trip shortly. I'm driving there, but I have my own car. The only other way is to catch the train to Madrid and then Madrid to Salamanca. The only problem with the train is when you get to Madrid you must not only change trains, you must also change Stations. The AVE does not go from Granada to Madrid.

Check with Renfe.com. You will find tickets and routes, there.
I think you need to go to Córdoba, then Madrid, then Salamanca, changing in those cities. But the website will give you the up to date information. |
