Do you have to go by car? It will take you around 4-5 hours. There is a high speed train that goes from Alicante.
Best road route is motorway, but it is a toll so you will pay.

It is about 4,5 hours. Look for the signs Valencia/Autovia del Este/Albacete. In the Area of Albacete you will have the signs indicatin Alicante. Keep the speed limit of 120 km/h. The fines are very high!!! for speeding. In many cases the radar controls are indicated, but not always! There is no toll road in between Madrid and Alicante, except you are taking the outer ring of Madrid. But thisone is quite cheap.
Besides, think in taking the train: www.renfe.es Fast, comfortable.Not yet high speed (above 250 km/h), but in it´s 200 km/h.
