My favourites:
The church "Sagrada Familia", Park Guell and the house of the architect Gaudi, there are also some buildings he designed that could be visited La pedrera and the Casa Batllo).
Just walking around in the streets of the old city (Barri Gotic). The old cathedral in the city centre.
The liberty quarter (Eixample)
The Aquarium is great and the Picasso Museum is a must.
Avoid las Ramblas: it's the main shopping area. It's not typical at all and very crowded an expensive.

The recommendations of Laura are OK.
If this is your first visit to the city I will recommended to take the touristic buses in front of El Corte Ingles at Plaza Cataluña. The Red and Blue lines can take you to the major attractions of the city like La Sagrada Familia, La Perdrera, Casa Batlo, Parque Guell (you have to walk up hill but is the park is beautiful), the Olympic city, the port with the aquarium, Barrio Gotico and La Rambla. I will also recommend to take the cremayera train up to El Tibidabo historical amusement park with the best view of the city. Las Ramblas can be crowded but don't miss the Mercado and the Colom monument. Walk carefully to the Contemporary Art Museum a splendid building by the American architect Richard Meier.

See any building constructed by Antonin Gaudi, he has a very interesting style. Go to the old gothic neighborhood. Visit Montjuic and everything around it. I disagree with the guy who said to avoid Las Ramblas. Yes, it's somewhat of a tourist trap, but you are a tourist aren't you? Just be careful of your personal belongings. The cathedral is enormous. Le Quatr Gats (The 4 Cats) is a restaurant in the old Gothic area that I highly recommend.
