The best of Madrid is the "juerga" (party, going out at night). The streets are always full of people and there are lots of bars. Spaniards go from bar to bar and they even have their beers on the streets (specially in the Summer, but all year round too).
Sunday mornings and afternoons are also fun, since everybody goes to have luch out, eating "tapas" in the "bares de tapas" everywhere, but specially in the "El Rastro" neighborhood where there's a flea market that day (oldest part of the city. Metro Station "La Latina"). Streets full of people again.

Besides the tourist sites suggested in other answers, I love the Plaza de Oriente, where the Royal Palace is located. Beautiful square (Metro station: Ópera).

In the Plaza Mayor there's a tourist office where you'll probably find a free newspaper called "In Madrid". It's in English and it has a lot of information of what's happening in the city.

The Prado,the Thyssen Bornemisza Museum, and the Reina Sofia Museum which features modern art. It also has the largest bull ring in the world and Estadio Santiago Bernabéu, the home to Real Madrid is also worth seeing- try and take in a game their before Beckham leaves.

If you have a free day to go outside Madrid, take the train to Segovia. The attached link has some of what you will see there.

There is a restaurant next to the Roman Aquiduct that serves the best roast suckling pig, too.

Definitely the Prado Museum, it closes early on Sundays, I think around noon. And Placa Mayor. and in between all that walking you will see so many other sites too!

The biggest must-see of Madrid; the hyper-galactic traffic jam and total collapse of the M-30 and M-40 circular highways every f****** day at peak hours. That's a real must see. Anyway, don't take your car with you 'cos you won't be able to park it. And I mean it. Residents only in most of the city. Anyone else has to pay and only for a very limited period of time. Great.

Cities!!! Granada, Toledo, Madrid are a must, because they are truely Spain. Depending on the time one has to travel in Spain.
Most will say Barcelona, but that is not true. Barcelona is Catalunia, and they let you know it, besides it is totally overpriced and tourist unfriendly.
Granada has the Alhambra, which is one of the most stunning sites in Europe. Granada and Toledo show much of Spains history. Get the Rough Guide or Lonely Planet Guide book for Spain. It helps.
