Cheapest will definitely be by bus, although probably longest too (around 19 hours). There is unlikely to be a direct route Barcelona-Gibraltar, so you should look for a bus to La LĂ­nea or Algeciras.

The cheapest and, almost certainly, quickest way to do this journey would be flying to Malaga, with a train connection to Fuengirola, and then bus to La Linea. You will be surprised at how cheap this can be done, as bus/train fares in Spain are very reasonable, whilst the flight is cheap too.
Veuling.com have 2 flights per day, 0615 & 1450 hrs, from Barcelona to Malaga. The journey is 1hr 30 mins and many flights are available at 25 euros inclusive of all taxes etc.
Trains leave from Malaga airport, at 13 mins & 43 mins past each hour, to Fuengirola, a journey of 29 minutes.
Buses from Fuengirola to La Linea leave at 0745hrs, 1100hrs, 1445hrs and 1745 hrs. You just walk across the border from La Linea into Gibraltar.

Check flights to the nearest airport, as well. They are pretty cheap, and would be much quicker than any other method. I don't know if there's an airport IN Gibraltar. If not, just take a bus from the nearest city (Cádiz?) that has one. This may NOT be your cheapest way, but you make it up by saving so much time.

I don't believe there is an actual train right into Gibraltar, and no matter what you do, you will wind up on a bus for some bit.

In my opinion it's not the cheapest you need worry about, it's finding the best way to do it. Driving is the best way if you are returning to Barcelona. If not driving I would get the AVE high speed train from Barcelona to Madrid, then the train from Madrid to Algeciras. From there you can get the bus to the Gibraltar border.
