Only one day?
The bus touristic is good but expensive.
Get up early. Get a taxi to the Sagrada Familia. Then a taxi to the Parc Guell. Then taxi to the Cathedral and explore the old part of town.
Lunch. Then walk up Paseo de Gracia and down Rambla de Catalunya.
By then you should need a siesta!

For only one day I strongly recommend the "Bus Turistic", the hop-on hop-off touristic bus.
Whenever I have received visits from abroad I have taken my guests to that bus because, apart from taking you to the most popular sights, you can also see districts or places that you would not see otherwise which are in between.
If your day in Barcelona happens to be when the local football team (soccer) FC Barcelona plays home, you should not miss a match at the huge Nou Camp stadium, even if you are not into football.
Do not ever think of attending a bullfight if it is Sunday. More than 90% of Catalans hate it, we are about to ban it and most of the attendance is of ignorant tourist who think they are going to see something typical in Barcelona, which in fact it is not.
