It's been said already, but have to reiterate:

STATUE OF LIBERTY.

Period.
She is the most beautiful woman that you will ever see on the face of this earth.

To add:
*Imagine mosaic at Strawberry Fields in Central Park
*Sunset from the Top of the Rock or ESB
*Circle Line boat tour (evening cruise is HIGHLY recommended)
*Reuben from Katz's
*Walk across the Brooklyn Bridge
*A shot of whiskey at the White Horse tavern where Dylan Thomas drank himself to death.
*Cheescake from Maxi's deli in Times Square (mmmm.....raspberry hazelnut chocolate)
*People watching at Rockefeller Plaza, City Hall Fountain, or any subway station

Since you have 5 days, which allows a lot of time to see quite a bit, I would recommend breaking up your days into neighborhoods (i.e. Downtown/LES/Chinatown/Little Italy one day, West Village/Chelsea/Tribeca/Meatpacking, Flatiron up to Midtown, Midtown/Central Park/Museums, UES/UWS/Harlem, etc)
If you plan your activities (or don't plan, which is how I did my last trip) by neighborhood, you wander upon the most specatular things that are remarkably un-touristy, full of history, interesting, tasty, or fun....and aren't in the travel guides!

Have fun!!!

The USS Intrepid Air and Space Museum. Look for the large gray ship onthe west side of Manhattan Island, between 40th and 42nd Streets (or is it 42nd and 44th?) with a bunch of aircraft on the top deck.

Someone else already suggested the Circle Line tour and the Statue of Liberty.

Coney Island.

The Ground Zero site.

Tiff.any.
