Subway. Go to http://www.mta.info/ and look up one-day passes.
They also have some tourism deals that include a full day Metrocard as well as discounted or free museum and tourist attraction admission.
Walking is unrealistic -- the city is massive and the sites are spread far apart. You'd spend most of your day walking around... which isn't actually a bad thing, unless you're looking to see the actual sites.

...basically everywhere we natives try to avoid ;). Well, except Central Park. We would all go crazy without Central Park. |

if you don't want to walk, take the subway. those are the cheapest ways of getting around. there's loads to do and see in NYC. museums and the zoo and Rockefeller Center and Macy's and art galleries. use this site to plan your trip. have a great time! |

An unlimited ride metrocard (for 1, 7, or 30 days) and you can go anywhere in New York City or even Long Island.
If you've got the time, you can go from the edge of Westchester in the Bronx out to the beaches in the Hamptons.
Mostly, stick to Manhattan, though. That's where most of the interesting things are. |

I've only been to Manhattan, but the best way to get around from a cost standpoint is to walk and take the subway. Definitely go to Times Square, Rockafeller Center, and the Statue of Liberty. There are a lot of museums and art galleries to see around the city. There are a lot of TV shows that give away free tickets too (shows like the Late Show will be booked unless you plan a long time in advance). I'd recommend going to some night clubs/jazz clubs as well.
For food, street vendors are cheap and many small local restaurants have good food for fairly low cost. |

Far and away - walking. Just don't think you're going to see the Cloisters and Ground Zero on the same day without taking the subway. Concentrate the areas you want to see. "Broadway?" Nothing much to see except the exterior of closed theaters. So what? Times Square is stricly for rubes, stay away. |

Subways will get you almost anywhere you want to go faster than buses or taxis. The unlimited ride tickets are a good deal.
As for what to see, I'd suggest you figure out what you want to experience and target only those destinations - most times you'll be confined in a smaller area and can get deeper into the experience. Examples:
Shopping - Fifth Avenue
Business - Wall Street, Midtown
Ethnic - Little Italy, Lower Second Ave, Chinatown, etc.
Artsy - SoHo, Greenwich Village
Skyline - Hoboken, NJ
Immigration - Lower Manhattan and the Statue of Liberty
Entertainment - Times Square and Broadway
Brooklyn - Brooklyn, Coney Island
Etc.
