I'm not sure which one is the best, but if you are looking for the CHEAPEST airline like you said, these are not in consideration:
- Garuda indonesia.
- Singapore airlines.
- Vietnam airlines.
Jetstar (Vietnam), Lion Air (Indonesia), Tiger Air (Singapore perhaps) ... are cheap Airlines. But because you fly from Singapore to Vietnam, it's better to pick Vietnam or Singapore Airline (not Indonesian). For Vietnamese people for instance, Jetstar sold very cheap tickets to fly in domestic or abroad.
For comfortable, it depends on what you'd like. For example: Foods, Seats ... If you go with Vietnamease Airlines for example (Jetstar Pacific, Vietnam Airline) then you can see the girls in Vietnamese traditional dress hehe ^_^ |

Its probably going to be a toss up between Air Asia and Jet star. Air asia just had a free ticket deal where you could get to Singapore for Taxes. About $45 USD. It was just a promotion though and the tickets available were for next april through June. Still, Air Asia is hard to beat. But... be aware that their weight restrictions on luggage are very low and you pay big bucks for overages. so make sure to read the fine print.

I used to stay in Singapore and travelled back to VN quite often. The cheapest and best quality is - I think - Tiger Airways. New air-plane and cheaper. I just bought a ticket from Singapore to Vietnam on Nov with 25kg luggage only 106SGD. So i think it is a good deal
