You can book a bus from HCMC to Siem Reap through your guesthouse/hotel or purchase the ticket yourself through any of the Sinh Cafe outlets that litter the Pham Ngu Lao street or backpackers area. There's also a SAPACO bus office in the smae street near or facing New World Hotel, just across the park.
I crossed the Bavet-Mocbai border several months (6 months) ago through the Sapaco bus booked by my guesthouse. Someone will pick you up at your guesthouse at around 6AM and take you to the bus. Crossing the border will take about 6 to 7 hours taking you to the capital of Phnom Penh, then you will wait for 30mins to 1 hour where you will change buses, one that will take you to Siem Reap - taking another 7-8 hours. You will be in SR by 9-9:30PM.
As far as I know, most of these buses will leave HCMC at 6AM.
