Flying is the easiest way and the Vietnam to Singapore leg its the only practical way to travel. Though if you reverse your plans and go from Vietnam to Thailand and then on to Singapore a land crossing is doable.
You can take a very nice private bus from Ho Chi Minh City to Phnom Penh, about 6 hours at a cost of approx. $6.00 or slower but more interesting way is to take a bus down to Chau Doc and a boat up the Mekong to Phnom Penh. From Phnom Penh to Siem Reap via either bus direct or boat across Lake Tonle Sap and then on to Bangkok via bus or an option would be to take a bus down to Sihanoukville from Phnom Penh. From Sihanoukvill a ferry or chartered boat up the coast to the Thai boarder city of Hat Lek and then another bus on to Bangkok. Once in Bangkok you take a train down through Southern Thailand, across Malaysia ending in Singapore. the mekong river is the only other way i know where you can legaly cross the borders... but there is plenty of jungle if you have other plans. there are plenty of tour agencyies there who can give you specifis and get you visas

The way recommend to you is going by bus.
From Bangkok you go to Phnom Penh (Cambodia) and then take another bus to Saigon (Vietnam). It cost total about 12 USD.
get your visa for Cambodia in Bangkok 20 USD.
First you get a bus from Bangkok to Siem riep (7 hour). Stay in there to visit Angkor Wat. and then take a bus to Phnom Penh. In Phnom Penh get visa to Vietnam 25 USD. take a bus to Saigon (viet nam) cost 5 USD and (6 hour by bus) you will arrive to Saigon Vietnam.
If you like to return Singapore you must go by plane. But you can get cheap tick in http://www.tigerairways.com.

From BKK to Vietnam is easily doable by land either through Cambodia or Laos. Both are way better countries than Thailand(IMHO)so it would be worth spending time in them as well. Going through Cambodia I prefer going the sea route from of Koh Kong rather than the in-land route via Poipet (the road is awful and Poipet is a pit, one of the only places in Cambodia you really have to look out for getting ripped off beware) but if you are going to ANgkor Wat, a must in if you are in the area it makes more sense. My recommendations are: get your visa for Cambodia in Bangkok 20$US or 25 if you get an agency to do it for you. Border is a rip of (for poipet) Take a bus only to the border and then arrange a taxi to Siem Reap with a few people. It is a bit more expensive but way faster and more comfortable. Bus ticket straight to Siem Reap is a rip off.
You have to get the Vietnamese visa in Phnom Penh from and agent for about 30$ you cannot get it on the border. THe roads in Cambodia are pretty good (other than from Poipet to Siem Reap) and bus prices are reasonable. Have fun it is an awesome country
